Coiled Therapeutics announces next-gen formulation of brain penetrant

(Sharecast News) - Coiled Therapeutics said on Tuesday that it had developed a next-generation lipid-based softgel formulation of AO-252, its first-in-class brain-penetrant TACC3 inhibitor, with first patient dosing expected in July 2026.

The AIM-traded company said the new formulation was designed to improve consistency and drug exposure at higher dose levels, addressing absorption limits seen with the current tablet.

It said the formulation supported its planned dose expansion cohorts in ovarian and prostate cancer, targeting enrolment of up to 30 patients by the fourth quarter.

Chief executive Sridhar Vempati said the softgel was "a key step in our dose expansion programme" and could "further strengthen AO-252's potential to deliver meaningful clinical outcomes."

At 0859 BST, shares in Coiled Therapeutics were up 4.74% at 9.95p.
